ABSTRACT
Aim/Background: To study the improvement effect of Phellinus linteus polysaccharides (Phps) on doxorubicin-induced myocarditis injury in mice. Materials and Methods: The mice were randomly divided into 5 groups, namely blank control group, model group, and Phps low-, medium-, high-dose groups. The model group and Phps groups were induced by doxorubicin to establish a myocardial injury model of mice. After 7 days, the physiological status of the mice was observed and the cardiac function indicators of each group (+dp/dtmax,-dp/dtmax,-dp/ dtmax, BP, EF, FS, HR) were measured. The activities of myocardial enzymes (AST, CK, LDH) were detected using a fully automated biochemical analyzer. The levels of inflammatory factors (IL-6, IL-1 β, TNF-α) in myocardial tissue were detected with ELISA kits. The pathological changes in myocardial tissue were detected using HE staining method. The mRNA expression levels of PI3K, Akt, and mTOR in myocardial tissue were detected using Real-time PCR. Results and Conclusion: Compared with the blank control group, the model group mice showed poorer states, with significant reductions in blood pressure, ejection fraction, and heart rate (p<0.05), the serum myocardial enzyme activity and inflammatory factor levels were significantly increased (p<0.05), and myocardial cells were thickened and swollen and deformed. Compared with the model group, the above indicators in treatment groups were effectively improved (p<0.05). Phps may mainly inhibit myocarditis through the PI3K/Akt/mTOR signaling pathway, thus effectively improving doxorubicin-induced myocardial injury in mice.
